Michael J. Fox, when addressing a comment concerning the idea brought up by this video that ammendment 2 would actually allow human cloning, responded thus:

"Well, I don't think that's true. You know, I campaigned for Claire McCaskill. And so I have to qualify it by saying I'm not qualified to speak on the page-to-page content of the initiative. Although, I am quite sure that I'll agree with it in spirit, I don't know, I -- On full disclosure, I haven't read it, and that's why I didn't put myself up for it distinctly."

Backing legislation he hasn't even read?!
Look out, America. It appears that Michael J. Fox is planning to run for Congress.

Role of the Pseudomonas aeruginosa dinB-Encoded DNA Polymerase IV in Mutagenesis.


Pseudomonas aeruginosa is a human opportunistic pathogen that chronically infects the lungs of cystic fibrosis patients and is the leading cause of morbidity and mortality of people afflicted with this disease. A striking correlation between mutagenesis and persistence of P. aeruginosa has been reported. In other well-studied organisms, error-prone replication by Y-family DNA polymerases contributes significantly to mutagenesis. Based on an analysis of the PAO1 genome sequence, P. aeruginosa contains a single Y-family DNA polymerase encoded by the dinB gene. As part of an effort to understand mechanisms of mutagenesis in P. aeruginosa we have cloned the P. aeruginosa dinB gene, and utilized a combination of genetic and biochemical approaches to characterize the activity and regulation of the P. aeruginosa DinB protein (Pa-DinB). Our results indicate that Pa-DinB is a distributive DNA polymerase that lacks intrinsic proofreading activity in vitro. Modest overexpression of Pa-DinB from a plasmid conferred a mutator phenotype in both E. coli and P. aeruginosa. Examination of this mutator phenotype indicated that Pa-DinB has a propensity to promote C-->A transversions, and -1 frameshift mutations within poly-dGMP and poly-dAMP runs. Characterization of lexA(+) and DeltalexA::aacC1 P. aeruginosa strains, together with in vitro DNA binding assays utilizing cell free extracts or purified P. aeruginosa LexA protein (Pa-LexA) indicated that transcription of the dinB gene is regulated as part of an SOS-like response. Deletion of the Pa-dinB gene sensitized P. aeruginosa to nitrofurazone and 4-nitroquinoline 1-oxide, consistent with a role for Pa-DinB in translesion DNA synthesis over N(2)-dG adducts. Finally, P. aeruginosa exhibited a UV-inducible mutator phenotype that was independent of Pa-dinB function, and instead required polA and polC, which encode DNA polymerases I and the second DNA polymerase III enzyme, respectively. Possible roles of the P. aeruginosa dinB, polA and polC gene products in mutagenesis are discussed.

Updatus Filmus
So, "The Prestige" is the film of the week... since it opened this weekend (and went straight to number one)...

But there are several others I'd like to draw your attention to, if I may...

"Pan's Labyrinth"
Guillermo del Toro, of Hellboy and Blade 2 fame, directs this international-buzz-producing fairy tale that looks, now that the full trailer has been released, wicked awesome.

"Climates" (aka, "Iklimler")
Turkish born Director/Actor Nuri Bilge Ceylan is already picking up massive reviews as the next great cinematic master. In this flick he totally hooks up with his real life wife, Ebru Ceylan.

"Jonestown"
Woof and a woof. Subtitled, "The Life and Death of Peoples Temple," this documentary tells the story of the 909 members of Peoples Temple, who followed Jim Jones all the way to the grave by way of the largest and most famous mass suicide of all time. Intense.

The End
Pan is over. Now that I have some time, I'll have to come up with some things to do...

Already on the to-do list:

- get a haircut.
I played a drunken pirate in the show. As a result, the director and costume designer wouldn't let me cut my hair (even a trim). As a result, my hair is now (as was desired) extremely shaggy, with innumerable split-ends, and dangerously approaching mullet status "in the back."

- dance back.
For those of you who don't know, I got served. Next step, contact a lawyer and figure out what needs to be included in a response letter so that I can be sure everything is correct and will be taken care of the first time around, as waiting these several months to get to this step (which was all of about 2.5 pages of legal BS, in triplicate) is something I hope to avoid in future steps.

- possibly updating this thing every once in a while.
But we all know how we feel about the holding of ones breath.

- get back to writing.
There are a few ideas I'm tossing around in the way of film scripts, both short and feature. Plus, I haven't written anything terribly comedic lately, so I'm considering forcing myself to throw together a t.v. pilot to see if it leads to anything interesting.
